The SUMO fusion system enhances soluble expression, promotes correct folding, and protects proteins from proteolytic degradation. SUMO protease specifically recognizes and cleaves the SUMO tag, allowing its complete removal with minimal impact on target protein structure and function.
DETAIBIO's His-tagged recombinant SUMO protease is expressed in E. coli and affinity-purified for convenient removal by Ni–NTA resin after cleavage.
Unit Definition
One unit (U) of SUMO protease cleaves >85% of 2 μg of a control SUMO-fusion substrate in 1 hour at 30 °C in 1× SUMO Protease Buffer (50 mM Tris–HCl, pH 8.0; 1 mM DTT).

Notes
- Use partially or fully purified SUMO-fusion substrate for optimal cleavage efficiency.
- NaCl concentration can be adjusted between 0–300 mM to accommodate different substrates.
